On Saturday, October 2, we organized a \u00ab\u00a0Visite Insolite\u00a0\u00bb with the CNRS, for the \u00ab\u00a0F\u00eate de la Science\u00a0\u00bb 30th anniversary. To offer a fun discovery of organoids, we mobilized around ten members from the Epigenetics and Cell Fate unit and the Jaques Monod Institute.<\/div>\n<\/div>\n

One visit, two groups and three workshops<\/h3>\n

The visit started with a short introduction given by Claire Rougeulle (coordinator of Labex Who Am I?, and deputy director of the Epigenetics and Cell Fate unit) on the concepts of stem cells, organoids and the CRISPR technology, recently awarded of a Nobel Prize.<\/p><\/div>\n

Then, the visitors left to discover the laboratories and equipment used by the researchers to study organoids.<\/span><\/p>\n

A first workshop was dedicated to\u00a0<\/span>the visit of a level 2 laboratory, and the discovery of stem cells<\/span><\/strong>. L\u00e9o Carrillo (Ph.D. student in the Epigenetics and Cell Fate unit) invited the participants to put on the required attire, a gown, and gloves, before entering the laboratory. L\u00e9o presented the different pieces of equipment: incubators, laminar flow cabinets, microscopes, etc., and materials necessary for the culture of stem cells before performing a demonstration of changing the nutritional medium for these cells. Then, he accompanied the participants in observing these stem cells under a microscope and in a practical workshop: changing the cell culture medium themselves!<\/span><\/p>\n<\/div>\n

For the second workshop, Kamal Bouhali (Research Engineer, enSCORE platform) and Mika\u00eblle Bocel (Research Engineer, enSCORE platform) guided our visitors to observe organoids at different stages of evolution<\/strong>. Using salt dough models and couscous or bulgur seeds, the participants followed the development of pseudo-neural organoids over time. Then, the team challenged them, objective: transfer the organoids, change the medium, and encapsulate them in a Matrigel matrix. Not easy!<\/div>\n

Finally, the third workshop was devoted to the observation of organoid sections under a fluorescence microscope<\/strong>. K\u00e9vin Daupin (Ph.D. student in the Epigenetics and Cellular Destiny Unit), Jad Saleh, and Matis Soleilhac (Ph.D. students at the Jacques Monod Institute) started by briefly explaining the concepts of immunofluorescence and microscopy. Then, they guided the participants through the multicolored observations of immunostained neural and intestinal organoids. All were amazed!<\/div>\n

Une visite, deux groupes et trois ateliers<\/h3>

La visite a commenc\u00e9 par une courte introduction donn\u00e9e par Claire Rougeulle (coordinatrice du Labex Who Am I? et directrice adjointe de l'unit\u00e9 \u00c9pig\u00e9n\u00e9tique et Destin Cellulaire) sur les concepts des cellules souches, des organo\u00efdes et de la technologie CRISPR r\u00e9cemment prim\u00e9e d\u2019un prix Nobel.<\/p>

Puis, les visiteurs, divis\u00e9s en petits groupes, sont partis \u00e0 la d\u00e9couverte des laboratoires et \u00e9quipements utilis\u00e9s par les chercheurs pour \u00e9tudier les organo\u00efdes :<\/p>

Un premier atelier \u00e9tait consacr\u00e9 \u00e0 la visite d'un laboratoire de niveau L2 et \u00e0 la d\u00e9couverte de cellules souches<\/strong>. L\u00e9o Carrillo (doctorant \u00e0 l'unit\u00e9 \u00c9pig\u00e9n\u00e9tique et Destin Cellulaire), a invit\u00e9 les participants \u00e0 enfiler la tenue de rigueur, une blouse et des gants, avant d'entrer dans le laboratoire. L\u00e9o a pr\u00e9sent\u00e9 les diff\u00e9rents \u00e9quipements : incubateurs, h\u00f4tes, microscopes, etc. et mat\u00e9riels n\u00e9cessaires \u00e0 la culture de cellules souches avant de r\u00e9aliser une d\u00e9monstration de changement de milieu nutritif pour ces cellules. Il a ensuite accompagn\u00e9 les participants dans l'observation de ces cellules souches au microscope puis dans un atelier pratique : changer eux-m\u00eame le milieu de culture cellulaire !<\/p>

Pour le deuxi\u00e8me atelier, ce sont Kamal Bouhali (Ing\u00e9nieur de recherche, plateforme enSCORE) et Mika\u00eblle Bocel (Ing\u00e9nieure d'\u00e9tudes, plateforme enSCORE) qui ont initi\u00e9 nos visiteurs du jour \u00e0 l'observation d'organoides \u00e0 diff\u00e9rents stades d'\u00e9volution<\/strong>. A l'aide de mod\u00e8les en p\u00e2te \u00e0 sel et diff\u00e9rents grains de couscous ou de boulgour, les participants ont pu observer l'\u00e9volution de pseudo-organo\u00efdes neuraux au cours du temps. Puis, un challenge leur a \u00e9t\u00e9 propos\u00e9 : transf\u00e9rer les organo\u00efdes, changer le milieu, et les encapsuler dans une matrice de Matrigel. Pas simple !<\/p>

Enfin, le troisi\u00e8me atelier \u00e9tait consacr\u00e9 \u00e0 l'observation de coupes d'organo\u00efdes au microscope \u00e0 fluorescence<\/strong>. K\u00e9vin Daupin (doctorant \u00e0 l'unit\u00e9 \u00c9pig\u00e9n\u00e9tique et Destin Cellulaire), Jad Saleh et Matis Soleilhac (doctorants \u00e0 l'Institut Jacques Monod), ont expliqu\u00e9 bri\u00e8vement les concepts d'immuno-fluorescence et de microscopie avant de guider les participants \u00e0 travers les observations multicolores d'organo\u00efdes neuraux et intestinaux immuno-marqu\u00e9s. Tous en ont pris plein les yeux !<\/p>
