Clostridium difficile<\/strong>,<\/em>\u00a0une bact\u00e9rie Gram positive, sporulante et ana\u00e9robie, est la cause majeure des diarrh\u00e9es post-antibiotiques et est responsable de la majorit\u00e9 des colites pseudomembraneuses.<\/p>\n

C\u2019est aujourd\u2019hui le principal ent\u00e9ropathog\u00e8ne bact\u00e9rien isol\u00e9 des diarrh\u00e9es nosocomiales chez l\u2019adulte. Les infections \u00e0 C. difficile sont dues g\u00e9n\u00e9ralement \u00e0 l\u2019ingestion de spores pr\u00e9sentes dans l\u2019environnement. La production de deux toxines (TcdA et TcdB) et leur r\u00e9gulation est un \u00e9l\u00e9ment d\u00e9terminant du pouvoir pathog\u00e8ne de cette bact\u00e9rie. Cependant d\u2019autres facteurs de virulence semblent intervenir au cours du cycle infectieux.<\/p>\n<\/div>\n<\/div>\n<\/div>\n

LES PRINCIPAUX OBJECTIFS DE NOS RECHERCHES SONT\u00a0:<\/h3>\n

– De d\u00e9finir les m\u00e9canismes r\u00e9gulant l\u2019expression des g\u00e8nes des toxines de C. difficile au cours du temps (phase de transition cellulaire) et en r\u00e9ponse \u00e0 certains signaux de l\u2019environnement (sources carbon\u00e9es, acides amin\u00e9s soufr\u00e9s), ainsi que leur transport \u00e0 travers la membrane bact\u00e9rienne<\/p>\n

– D\u2019identifier les autres propri\u00e9t\u00e9s bact\u00e9riennes intervenant dans le processus de pathog\u00e9n\u00e8se de C. difficile incluant les facteurs de la colonisation du tube digestif de l\u2019h\u00f4te, les biofilms et la sporulation<\/p>\n

– D\u2019identifier les facteurs responsables du caract\u00e8re \u00e9mergent et hypervirulent des souches \u00e9pid\u00e9miques r\u00e9centes isol\u00e9es tant aux Etats-Unis qu\u2019en Europe<\/p>\n<\/div>\n<\/div>\n<\/div>\n
