L\u2019arthrose est la maladie articulaire la plus r\u00e9pandue, l\u2019invalidit\u00e9 qu\u2019elle induit par les douleurs et une perte de mobilit\u00e9. Elle se caract\u00e9rise par la destruction du cartilage secondaire \u00e0 une mort du chondrocyte, la seule cellule qui constitue le cartilage articulaire. En d\u00e9pit des efforts nombreux, les traitements visent actuellement \u00e0 r\u00e9duire les sympt\u00f4mes, aucun traitement sp\u00e9cifique n\u2019est disponible. Les l\u00e9sions \u00e9tant irr\u00e9versibles, le d\u00e9fi d\u2019une th\u00e9rapie efficace repose sur l\u2019incapacit\u00e9 du cartilage \u00e0 pouvoir se r\u00e9parer.<\/p>\n

L’ unit\u00e9 de recherche Bioscar (Universit\u00e9 Paris Cit\u00e9 et Inserm), s’est int\u00e9ress\u00e9e \u00e0 Lin28a, une prot\u00e9ine inhibitrice des MiRNA de la famille des Let-7, et dont l\u2019expression est associ\u00e9e \u00e0 la r\u00e9g\u00e9n\u00e9ration de nombreux tissus. Grace \u00e0 un mod\u00e8le murin d\u2019arthrose, l’\u00e9quipe de recherche a montr\u00e9 que la d\u00e9l\u00e9tion de Lin28a sp\u00e9cifiquement chondrocytaire aggrave la d\u00e9gradation du cartilage. En revanche, la surexpression de Lin28a uniquement dans le cartilage conduit \u00e0 maintenir un cartilage sain avec l\u2019\u00e2ge, et aussi la progression de l\u2019arthrose en activant les capacit\u00e9s de \u00a0reprogrammation des chondrocytes arthrosiques en chondrocytes sains. Ces r\u00e9sultats sugg\u00e8rent que Lin28a est naturellement r\u00e9-exprim\u00e9 par le cartilage arthrosique de fa\u00e7on \u00e0 limiter la d\u00e9gradation du cartilage.<\/p>\n

Cette \u00e9tude incite \u00e0 faire sur-exprimer directement Lin28a par le chondrocyte pour induire la r\u00e9g\u00e9n\u00e9ration du cartilage arthrosique chez l\u2019Homme. Les professeurs Martine Cohen Solal et Eric Hay du laboratoire BIOSCAR pr\u00e9voient de poursuivre leurs travaux en ciblant les MiRNA Let-7b et c. Les nouveaux vecteurs intelligents, qui permettent d’administrer des petits ARN avec une grande sp\u00e9cificit\u00e9 tissulaire, pourraient \u00eatre d\u00e9velopp\u00e9s pour limiter l’effet inhibiteur du MiRNA Let-7 sp\u00e9cifiquement dans le cartilage et promouvoir la reprogrammation des chondrocytes. Cette strat\u00e9gie est cliniquement pertinente puisque qu\u2019elle aurait l\u2019avantage de bloquer le d\u00e9veloppement de la maladie chez des patients symptomatiques aux stades pr\u00e9coces de la maladie.<\/p>\n
