z_index_tablet=\u00a0\u00bb500″]<\/p>\n La maladie r\u00e9nale chronique (MRC) est caract\u00e9ris\u00e9e par une perte progressive de la fonction des n\u00e9phrons, les unit\u00e9s fonctionnelles du rein. Les n\u00e9phrons sains restants peuvent, dans les premi\u00e8res phases de la maladie, prendre en charge le travail suppl\u00e9mentaire. Toutefois, dans le temps, ce surtravail endommage les n\u00e9phrons restants ce qui peut conduire \u00e0 la perte compl\u00e8te de la fonction r\u00e9nale. Sans fonction r\u00e9nale on ne peut pas survivre. Les patients qui sont dans cette situation doivent recourir \u00e0 la dialyse et\/ou \u00e0 la transplantation r\u00e9nale.<\/p>\n La vitesse de progression vers l\u2019insuffisance r\u00e9nale terminale est variable d\u2019un individu \u00e0 l\u2019autre. Il est donc n\u00e9cessaire d\u2019identifier des pr\u00e9dicteurs (bio-marqueurs) capables de pr\u00e9dire l\u2019\u00e9volution de la maladie et donc d\u2019identifier les patients qui pourraient b\u00e9n\u00e9ficier le mieux d\u2019une th\u00e9rapie. L\u2019\u00e9quipe de Fabiola Terzi<\/strong> vise \u00e0 mieux comprendre les m\u00e9canismes \u00e0 l\u2019origine de la destruction des n\u00e9phrons lors d\u2019une r\u00e9duction n\u00e9phronique. Ses travaux ont montr\u00e9 le r\u00f4le cl\u00e9 jou\u00e9 dans le processus l\u00e9sionnel d\u2019une voie mol\u00e9culaire, appel\u00e9e la voie de l\u2019EGFR (Epidermal Growth Factor Receptor<\/em>). L\u2019\u00e9quipe vient d\u2019identifier un nouveau m\u00e9dicament capable de bloquer un acteur cl\u00e9 de cette voie et donc le processus l\u00e9sionnel. D\u2019autre part, la mesure de certaines mol\u00e9cules de cette voie dans les urines semble capable de pr\u00e9dire l\u2019\u00e9volution de la maladie.<\/p>\n L\u2019\u00e9quipe de Marco Pontoglio<\/strong> s\u2019int\u00e9resse \u00e0 une maladie monogenique (l\u2019haploinsufficance de HNF1B) qui peut se manifester, entre autres, avec une MRC tr\u00e8s s\u00e9v\u00e8re chez l\u2019enfant. L\u2019\u00e9quipe vise \u00e0 identifier des nouveaux g\u00e8nes capables de contr\u00f4ler l\u2019\u00e9volution de la maladie et d\u2019expliquer pourquoi, pour une m\u00eame mutation, certains patients d\u00e9veloppent une maladie s\u00e9v\u00e8re, alors que d\u2019autre sont prot\u00e9g\u00e9s.
