Infirmi\u00e8re et titulaire d\u2019un master en sant\u00e9 publique sp\u00e9cialis\u00e9 dans les risques environnementaux, H\u00e9l\u00e8ne Amazouz a effectu\u00e9 sa th\u00e8se d\u2019\u00e9pid\u00e9miologie sous la direction du Pr Isabelle Momas et du Dr Fanny Ranci\u00e8re au sein de l\u2019\u00e9quipe HERA, du CRESS (U1153, Inserm) \u00e0 la facult\u00e9 de Pharmacie d’Universit\u00e9 Paris cit\u00e9.
Elle est actuellement post-doctorante en \u00e9pid\u00e9miologie au sein de l\u2019\u00e9quipe \u00ab Exposome & H\u00e9r\u00e9dit\u00e9 \u00bb du CESP (U1018, Inserm), gr\u00e2ce au soutien de la Fondation pour la recherche m\u00e9dicale (FRM). Son projet durant 3 ans porte sur l\u2019\u00e9tude des relations entre les expositions environnementales\/l\u2019alimentation et le risque d\u2019endom\u00e9triose dans les cohortes Constances et NutriNet-Sant\u00e9.<\/p>\n

Au cours des derni\u00e8res d\u00e9cennies, la pr\u00e9valence des pathologies allergiques a fortement augment\u00e9. Plusieurs facteurs (h\u00e9r\u00e9ditaires, environnementaux et comportementaux) peuvent influencer cette augmentation et notamment l\u2019alimentation qui a largement \u00e9volu\u00e9e ces derni\u00e8res ann\u00e9es (d\u00e9veloppement de laits infantiles, r\u00e9gime occidental, aliments ultra transform\u00e9s, etc.). Son r\u00f4le dans les pathologies allergiques et respiratoires reste peu \u00e9tudi\u00e9, ou incertain comme c\u2019est le cas pour l\u2019allaitement maternel. Le r\u00e9gime m\u00e9diterran\u00e9en riche notamment en antioxydants pourrait avoir des effets b\u00e9n\u00e9fiques sur la sant\u00e9 respiratoire et les allergies au cours de l\u2019enfance. De plus, l\u2019alimentation est un facteur de risque environnemental pertinent car il est dit modifiable, c\u2019est-\u00e0-dire qu\u2019il est possible de mettre en place des interventions pour influencer ou modifier certaines pratiques alimentaires au sein d\u2019une population.<\/p>\n

Dans ce contexte, mes travaux de recherches en sant\u00e9 publique ont vis\u00e9 \u00e0 explorer, pour la premi\u00e8re fois dans la cohorte PARIS, les donn\u00e9es d\u2019alimentation du nourrisson et de l\u2019enfant \u00e0 8 ans. Mes objectifs ont \u00e9t\u00e9 (1) d\u2019identifier et de caract\u00e9riser des profils\/groupes d\u2019alimentation (du nourrisson et de l\u2019enfant) et (2) d\u2019\u00e9tudier les liens avec la sant\u00e9 respiratoire\/les allergies chez les enfants.<\/p>\n

Mes travaux de th\u00e8se ont port\u00e9 sur des donn\u00e9es de grande qualit\u00e9 issues du suivi des 3840 enfants franciliens de la cohorte de naissances PARIS, \u00e9tude \u00e9pid\u00e9miologique prospective men\u00e9e en population g\u00e9n\u00e9rale. Le suivi des enfants a eu lieu r\u00e9guli\u00e8rement gr\u00e2ce \u00e0 des questionnaires et \u00e0 des bilans de sant\u00e9 clinico-biologiques effectu\u00e9s dans des centres de r\u00e9f\u00e9rence en allergologie p\u00e9diatrique (Necker et Trousseau, AP-HP). \u00c0 partir de ces donn\u00e9es, j\u2019ai dans un premier temps construit des profils d\u2019alimentation pr\u00e9coce (1\u00e8re ann\u00e9e de vie) gr\u00e2ce \u00e0 une m\u00e9thode statistique de regroupement non supervis\u00e9e multidimensionnelle et longitudinale (analyse de cluster k-means longitudinale). Dans un second temps, des groupes d\u2019alimentation \u00e0 8 ans ont \u00e9galement \u00e9t\u00e9 construits gr\u00e2ce \u00e0 la m\u00e9thode k-means et j\u2019ai calcul\u00e9 deux scores d\u2019adh\u00e9sion au r\u00e9gime m\u00e9diterran\u00e9en \u00e0 l\u2019\u00e2ge de 8 ans. J\u2019ai ensuite \u00e9tudi\u00e9 les associations entre ces profils d\u2019alimentation et la morbidit\u00e9 allergique\/respiratoire \u00e0 l\u2019aide de mod\u00e8les de r\u00e9gression lin\u00e9aire et logistique ajust\u00e9s sur les facteurs de confusions potentiels.<\/p>\n

Les principaux r\u00e9sultats de mes travaux de th\u00e8se montrent que l\u2019alimentation des nourrissons et des enfants de la cohorte PARIS est associ\u00e9e \u00e0 la sant\u00e9 respiratoire et aux allergies \u00e0 8 ans. Compar\u00e9s aux enfants caract\u00e9ris\u00e9s par la prise de pr\u00e9parations infantiles standards, les enfants du profil \u00ab allaitement \u00bb tendaient \u00e0 avoir un risque plus faible d\u2019asthme. De plus, les enfants du profil \u00ab pr\u00e9parations infantiles partiellement hydrolys\u00e9es avec un label hypoallerg\u00e9nique \u00bb avaient une augmentation du risque d\u2019\u00eatre sensibilis\u00e9s \u00e0 au moins un allerg\u00e8ne, d\u2019avoir une FeNO >20 ppb (marqueur d\u2019inflammation bronchique) et une diminution des param\u00e8tres de la fonction respiratoire (VEMS, CVF) \u00e0 8 ans. S\u2019agissant de l\u2019alimentation durant l\u2019enfance, une alimentation \u00ab plus saine \u00bb ou \u00ab plus diversifi\u00e9e \u00bb ou une forte adh\u00e9sion au r\u00e9gime m\u00e9diterran\u00e9en \u00e0 8 ans \u00e9tait b\u00e9n\u00e9fique pour la sant\u00e9 respiratoire, l\u2019asthme et la sensibilisation allerg\u00e9nique.<\/p>\n

Mes travaux contribuent \u00e0 am\u00e9liorer les connaissances sur les liens entre alimentation de l\u2019enfant et sant\u00e9 respiratoire\/allergies. Les r\u00e9sultats soutiennent les recommandations actuelles de sant\u00e9 publique privil\u00e9giant l\u2019allaitement et une alimentation saine durant l\u2019enfance, et apportent de nouvelles pistes pour pr\u00e9venir les probl\u00e8mes respiratoires et allergiques par l\u2019alimentation. Mes r\u00e9sultats de recherche de th\u00e8se ont \u00e9t\u00e9 pr\u00e9sent\u00e9s sous forme de communications orales et \u00e9crites dans des congr\u00e8s internationaux et nationaux. Ils sont \u00e9galement publi\u00e9s sous forme d\u2019articles originaux dans des revues internationales de rang A, Allergy, Pediatric Allergy and Immunology<\/em>, et un troisi\u00e8me est en cours de r\u00e9vision.<\/p>\n

En conclusion, mes travaux de recherche en \u00e9pid\u00e9miologie environnementale ont apport\u00e9 de nouvelles connaissances et ont fait progresser la science sur un sujet d\u2019int\u00e9r\u00eat majeur pour la sant\u00e9 publique. Dans mon manuscrit de th\u00e8se, j\u2019ai pris plaisir \u00e0 d\u00e9velopper attentivement une partie montrant l\u2019int\u00e9r\u00eat en sant\u00e9 publique de mes travaux de th\u00e8se et \u00e0 analyser leurs potentielles retomb\u00e9es en termes d\u2019aide \u00e0 la prise de d\u00e9cision. En effet, les recherches men\u00e9es dans le cadre de mon projet doctoral peuvent orienter les d\u00e9cideurs vers diverses actions \u00e0 mettre en \u0153uvre afin d\u2019am\u00e9liorer l\u2019\u00e9tat de sant\u00e9 des populations au regard des pathologies respiratoires et allergiques p\u00e9diatriques, notamment via l\u2019alimentation. De plus, ces travaux de th\u00e8se montrent que pour informer le grand public des recommandations de sant\u00e9 publique, le r\u00f4le des professionnels de sant\u00e9 est primordial. Les pharmaciens d\u2019officine sont un \u00e9l\u00e9ment cl\u00e9 du syst\u00e8me de sant\u00e9 fran\u00e7ais. Gr\u00e2ce \u00e0 leur expertise et \u00e0 la relation de confiance soignant\/soign\u00e9 et de proximit\u00e9 qu\u2019ils entretiennent avec les patients, ils sont des acteurs de choix pour diffuser les messages de pr\u00e9vention cl\u00e9s (ex : informations sur l\u2019allaitement, conseils sur le choix des pr\u00e9parations pour nourrissons, etc.).<\/p>\n

Sa recherche est principalement dirig\u00e9e sur la pr\u00e9disposition g\u00e9n\u00e9tique et immunologique aux infections virales s\u00e9v\u00e8res, notamment les causes et cons\u00e9quences des auto-anticorps anti-interf\u00e9rons de type I. Au cours de son master 2 puis de sa th\u00e8se, il a \u00e9tudi\u00e9 la variabilit\u00e9 clinique inter-individuelle au cours d\u2019une infection virale. Dans l\u2019\u00e9quipe de Jean-Laurent Casanova, il a travaill\u00e9 sur plusieurs variants dans le g\u00e8ne IFNAR1 conf\u00e9rant une pr\u00e9disposition \u00e0 faire des infections virales particuli\u00e8rement s\u00e9v\u00e8res, notamment l\u2019enc\u00e9phalite herp\u00e9tique, les r\u00e9actions s\u00e9v\u00e8res au vaccin vivant de la fi\u00e8vre jaune et les pneumopathies \u00e0 COVID-19 s\u00e9v\u00e8re. Ceci d\u00e9montre que des formes s\u00e9v\u00e8res de COVID-19 pouvaient \u00eatre dues \u00e0 un d\u00e9faut de r\u00e9ponse de la voie de l\u2019interf\u00e9ron de type I. Cette de\u0301couverte a conduits a\u0300 tester l\u2019hypothe\u0300se selon laquelle une pneumonie COVID-19 grave pourrait re\u0301sulter d\u2019une atteinte de l\u2019immunite\u0301 de\u0301pendante des IFN de type I, mais re\u0301sultant d\u2019autres causes chez les patients n\u2019ayant pas ces de\u0301fauts ge\u0301ne\u0301tiques, selon l\u2019hypothe\u0300se que la neutralisation des auto-Ac contre l\u2019IFN de type I agirait comme une phe\u0301nocopie auto-immune des de\u0301fauts ge\u0301ne\u0301tiques de l\u2019immunite\u0301 lie\u0301e a\u0300 l\u2019IFN de type I et pourrait donc e\u0302tre a\u0300 l\u2019origine d\u2019un COVID-19 potentiellement mortel. Son travail, et celui de nombreux autres coll\u00e8gues et collaborateurs a montr\u00e9 que les auto-Ac neutralisant les IFNs de type I pr\u00e9c\u00e8dent l\u2019infection \u00e0 SARS-CoV-2 et les autres infections virales, augmentent de fa\u00e7on importante apr\u00e8s 70 ans, et expliquent au moins 20% des formes s\u00e9v\u00e8res de COVID-19 chez les individus de plus de 80 ans et environ 20% des formes fatales \u00e0 tout \u00e2ge. Il semble assez ironique que les formes se\u0301ve\u0300res de COVID-19 puissent re\u0301sulter d\u2019une attaque auto-immune et adaptative contre l\u2019immunite\u0301 intrinse\u0300que et inne\u0301e. Au total, ces d\u00e9couvertes ont permis de d\u00e9montrer que les IFNs de type I sont essentiels dans la lutte anti-SARS-CoV-2 et qu\u2019un d\u00e9faut (g\u00e9n\u00e9tique ou auto-immun) pr\u00e9dispose \u00e0 faire une forme s\u00e9v\u00e8re. Les implications diagnostiques et th\u00e9rapeutiques sont imm\u00e9diates et importantes pour les patients.<\/p>\n

En tant que m\u00e9decin interne en cours de sp\u00e9cialisation en h\u00e9matologie aux H\u00f4pitaux Universitaires de Gen\u00e8ve, cette th\u00e8se s\u2019est naturellement invit\u00e9e dans le parcours de formation de Laetitia. C\u2019est lors de sonprojet de master en 5\u00e8me<\/sup> ann\u00e9e de m\u00e9decine qu’elle a \u00e9t\u00e9 fascin\u00e9e par l\u2019allogreffe de cellules souches h\u00e9matopo\u00ef\u00e9tiques, une th\u00e9rapie au croisement de l\u2019immunologie et l\u2019oncoh\u00e9matologie. Il s\u2019agit d\u2019une discipline tr\u00e8s complexe aux enjeux croissants pour la recherche fondamentale et translationnelle.
\u00ab\u00a0Je suis tr\u00e8s heureuse d\u2019avoir accompli cette th\u00e8se de doctorat, repr\u00e9sentant un r\u00e9el d\u00e9fi scientifique pour l\u2019h\u00e9matologie et je me r\u00e9jouis de poursuivre mon chemin de m\u00e9decin chercheur dans ce domaine passionnant<\/em>\u00ab\u00a0.

\u00a0\u00a0\u00a0\u00a0 La rupture de tol\u00e9rance immunitaire est \u00e0 l’origine des ph\u00e9nom\u00e8nes d’auto-immunit\u00e9, du rejet de greffe d’organe et de la maladie du greffon contre l’h\u00f4te (GvHD) dans le cadre d\u2019une allogreffe de cellules souches h\u00e9matopo\u00ef\u00e9tiques (CSH). Une telle greffe repr\u00e9sente une th\u00e9rapie curative essentielle pour de nombreuses pathologies h\u00e9matologiques acquises et cong\u00e9nitales. Toutefois, atteindre un \u00e9tat de tol\u00e9rance apr\u00e8s un tel traitement reste un d\u00e9fi majeur pour les cliniciens afin d\u2019\u00e9viter l’effet d\u00e9vastateur de la GvHD. Suite \u00e0 une allogreffe de CSH, certains patients d\u00e9velopperont un \u00e9tat de tol\u00e9rance immunitaire sous aucune immunosuppression, une condition clinique nomm\u00e9e tol\u00e9rance op\u00e9rationnelle. Ces receveurs ont un syst\u00e8me immunitaire pleinement fonctionnel, capable de pr\u00e9venir les rechutes h\u00e9matologiques et de contr\u00f4ler les infections. Les m\u00e9canismes biologiques sous-jacents \u00e0 cet \u00e9tat impliquent probablement \u00e0 la fois des m\u00e9canismes centraux et p\u00e9riph\u00e9riques, mais ces derniers restent encore mal compris, emp\u00eachant le d\u00e9veloppement de th\u00e9rapies cibl\u00e9es. La r\u00e9ponse immunitaire \u00e9tant un processus hautement dynamique r\u00e9sultant d’interactions complexes entre les cellules immunitaires et leur microenvironnement, nous avons \u00e9mis l’hypoth\u00e8se que la tol\u00e9rance op\u00e9rationnelle \u00e9merge d’une adaptation du syst\u00e8me immunitaire dans sa globalit\u00e9 apr\u00e8s la greffe.<\/p>\n

\u00a0\u00a0\u00a0\u00a0 \u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 Au sein de deux cohortes ind\u00e9pendantes de patients ayant b\u00e9n\u00e9fici\u00e9 d’une allogreffe de CSH \u00e0 partir d’un donneur HLA-identique apparent\u00e9, nous avons collect\u00e9 des \u00e9chantillons de sang chez les receveurs 1 \u00e0 2 ans apr\u00e8s la transplantation ainsi que chez leurs donneurs avant la collection de CSH. Nous avons \u00e9tudi\u00e9 via des outils mutiomiques leurs profils ph\u00e9notypiques immunitaires, transcriptomiques et m\u00e9tabolomiques avant de mettre en \u0153uvre une analyse int\u00e9grative des donn\u00e9es incluant les variables cliniques.<\/p>\n

\u00a0\u00a0\u00a0\u00a0 \u00a0\u00a0\u00a0\u00a0\u00a0\u00a0 Ces analyses int\u00e9gratives ont r\u00e9v\u00e9l\u00e9 un r\u00f4le central de l’ectoenzyme CD38 corr\u00e9l\u00e9e \u00e0 l\u2019activation du syst\u00e8me compl\u00e9ment et des plaquettes, dans la r\u00e9ponse allo-immune persistante chez les patients non tol\u00e9rants (avec GvHD sous immunosuppresseurs). En parall\u00e8le, les patients tol\u00e9rants pr\u00e9sentaient des st\u00e9ro\u00efdes androg\u00e8niques plus \u00e9lev\u00e9s, associ\u00e9s \u00e0 un r\u00e9seau immunitaire caract\u00e9ris\u00e9 par des CD8 + na\u00effs et des lymphocytes T doubles n\u00e9gatifs avec un profil transcriptomique impliquant le catabolisme des nucl\u00e9otides par NT5E \/ CD73. Les fonctions oppos\u00e9es des ectoenzymes CD38 et CD73 dans la signalisation des purines et la r\u00e9gulation immunitaire pourraient expliquer la balance entre un \u00e9tat non tol\u00e9rant et un \u00e9tat tol\u00e9rant apr\u00e8s l\u2019allogreffe de CSH. La production d’ad\u00e9nosine li\u00e9e au CD73 semble \u00eatre un \u00e9l\u00e9ment r\u00e9gulateur cl\u00e9 de la tol\u00e9rance op\u00e9rationnelle. Bien que ces d\u00e9couvertes doivent \u00eatre confirm\u00e9es par des analyses fonctionnelles, cette \u00e9tude ouvre de nouvelles portes dans la compr\u00e9hension de la tol\u00e9rance immunitaire apr\u00e8s l\u2019allogreffe de CSH avec des cibles th\u00e9rapeutiques potentielles qui pourraient am\u00e9liorer les r\u00e9sultats en clinique des patients non tol\u00e9rants.<\/p>\n

Apr\u00e8s avoir obtenu son baccalaur\u00e9at scientifique au Lyc\u00e9e Marguerite de Flandre (Gondecourt, Nord), J\u00e9r\u00e9my Dufloo a int\u00e9gr\u00e9 une classe pr\u00e9paratoire BCPST au Lyc\u00e9e Henri IV (Paris). Au terme de ces deux ann\u00e9es, il a \u00e9t\u00e9 admis \u00e0 l\u2019\u00c9cole Normale Sup\u00e9rieure de Lyon o\u00f9 il a obtenu sa Licence et mon Master \u00ab Biosciences \u00bb.
Au cours de ses \u00e9tudes sup\u00e9rieures, J\u00e9r\u00e9my a r\u00e9alis\u00e9 plusieurs stages de recherche en France (Centre International de Recherche en Infectiologie, Lyon) et \u00e0 l\u2019\u00e9tranger (Doherty Institute, Melbourne, Australie ; Ragon Institute of MGH, MIT and Harvard, Boston, Etats-Unis) pour \u00e9tudier divers aspects de l\u2019\u2019interaction h\u00f4te-VIH-1. Il a ensuite rejoint l\u2019Institut Pasteur et l\u2019unit\u00e9 \u00ab Virus et Immunit\u00e9 \u00bb pour y r\u00e9aliser sa th\u00e8se sous la direction du Pr Olivier Schwartz et la co-supervision du Dr Timoth\u00e9e Bruel. Ses travaux de doctorat, financ\u00e9s par le Minist\u00e8re de l\u2019Enseignement sup\u00e9rieur et de la Recherche, portaient sur la caract\u00e9risation des activit\u00e9s antivirales des anticorps g\u00e9n\u00e9r\u00e9s lors d\u2019une infection par le VIH-1 ou le SARS-CoV-2. Il est actuellement chercheur postdoctoral dans le laboratoire du Dr Rafael Sanju\u00e1n \u00e0 l\u2019Universit\u00e9 de Valencia (Espagne), financ\u00e9 par l\u2019Organisation europ\u00e9enne de biologie mol\u00e9culaire (EMBO). Il \u00e9tudie exp\u00e9rimentalement les risques d\u2019\u00e9mergence de nouveaux pathog\u00e8nes viraux.<\/p>\n

Les anticorps produits lors d\u2019une infection virale ont de nombreuses fonctions. Ils se fixent aux virions et emp\u00eachent l\u2019infection des cellules cibles, un processus nomm\u00e9 neutralisation. Ils peuvent \u00e9galement se lier aux particules virales ou aux cellules infect\u00e9es et recruter d\u2019autres effecteurs immunitaires, comme le compl\u00e9ment ou les cellules Natural Killer<\/em> (NK). Dans cette th\u00e8se, nous avons caract\u00e9ris\u00e9 les activit\u00e9s antivirales des anticorps induits lors d\u2019une infection par le VIH-1 ou par le SARS-CoV-2.<\/p>\n

Certains patients infect\u00e9s par le VIH-1 d\u00e9veloppent des anticorps efficaces contre la majorit\u00e9 des souches virales circulantes. Ces anticorps sont d\u00e9nomm\u00e9s anticorps neutralisants \u00e0 large spectre, ou bNAbs (broadly neutralizing antibodies<\/em>). La plupart des strat\u00e9gies vaccinales visent \u00e0 induire la production de ces bNAbs. Ils sont \u00e9galement des candidats de choix pour des approches th\u00e9rapeutiques innovantes. Leur large spectre de neutralisation ainsi que leur capacit\u00e9 \u00e0 induire la lyse des cellules infect\u00e9es apr\u00e8s activation des cellules NK ont \u00e9t\u00e9 largement caract\u00e9ris\u00e9s. Dans ce travail de th\u00e8se, nous avons caract\u00e9ris\u00e9 deux nouvelles activit\u00e9s antivirales des bNAbs. Dans une premi\u00e8re \u00e9tude, nous avons d\u00e9montr\u00e9 que les bNAbs sont capables d\u2019activer le compl\u00e9ment \u00e0 la surface de lymphocytes T CD4 infect\u00e9s par le VIH-1. Cette activation est sous-optimale et ne permet pas la lyse des cellules infect\u00e9es. La mort cellulaire d\u00e9pendante du compl\u00e9ment est inhib\u00e9e par la prot\u00e9ine r\u00e9gulatrice CD59 et par les prot\u00e9ines virales accessoires Vpu et Nef. Malgr\u00e9 cette absence de lyse rapide des cellules infect\u00e9es, l\u2019activation du compl\u00e9ment semble diminuer leur survie \u00e0 long terme. Dans une seconde \u00e9tude, nous avons d\u00e9montr\u00e9 que les bNAbs emp\u00eachent la lib\u00e9ration des particules virales depuis des cellules infect\u00e9es par le VIH-1. Les bNAbs forment \u00e0 la surface des cellules infect\u00e9es de larges complexes immuns de virions. Cette activit\u00e9 requiert la bivalence des bNAbs et n\u2019est observ\u00e9e qu\u2019avec les anticorps les plus puissants.<\/p>\n

Nous nous sommes \u00e9galement int\u00e9ress\u00e9s \u00e0 la r\u00e9ponse humorale d\u00e9clench\u00e9e lors d\u2019une infection par le SARS-CoV-2. Nous avons montr\u00e9 que les anticorps de patients infect\u00e9s par le SARS-CoV-2 d\u00e9clenchent une activation sous-optimale du compl\u00e9ment \u00e0 la surface des cellules infect\u00e9es n\u2019aboutissant pas \u00e0 la mort cellulaire. En revanche, les anticorps anti-SARS-CoV-2 peuvent induire la mort des cellules infect\u00e9es en recrutant les cellules NK. Enfin, nous avons compar\u00e9 la fonctionnalit\u00e9 des anticorps g\u00e9n\u00e9r\u00e9s lors d\u2019une infection symptomatique ou asymptomatique par le SARS-CoV-2, ce dernier cas concernant pr\u00e8s de la moiti\u00e9 des individus infect\u00e9s. Dans les deux cas, la r\u00e9ponse humorale g\u00e9n\u00e9r\u00e9e est polyfonctionnelle et capable \u00e0 la fois de neutraliser le virus et de cibler les cellules infect\u00e9es en activant le compl\u00e9ment ou les cellules NK. Les patients symptomatiques ont des niveaux d\u2019anticorps l\u00e9g\u00e8rement plus \u00e9lev\u00e9s, et en cons\u00e9quence une r\u00e9ponse humorale l\u00e9g\u00e8rement plus fonctionnelle.<\/p>\n

Ces r\u00e9sultats d\u00e9montrent que les activit\u00e9s des anticorps anti-VIH-1 et anti-SARS-CoV-2 ne se limitent pas \u00e0 la neutralisation. Plusieurs fonctions, chacune agissant de mani\u00e8re diff\u00e9rente sur les cellules infect\u00e9es et les particules virales, participent \u00e0 l\u2019activit\u00e9 antivirale des anticorps. Comprendre leurs interactions permettra de d\u00e9terminer les modes d\u2019actions des anticorps th\u00e9rapeutiques et vaccinaux et d\u2019optimiser leur utilisation ou leur induction.<\/p>\n

Jonas Landman a int\u00e9gr\u00e9 l’Ecole Polytechnique en 2013, puis a \u00e9tudi\u00e9 \u00e0 l’Universit\u00e9 de Berkeley en Californie en 2017. Son parcours scientifique g\u00e9n\u00e9raliste m\u00eale alors Math\u00e9matiques, Physique, Machine Learning, mais aussi Biologie, Economie ou Musique. Lors d’une conf\u00e9rence \u00e0 la NASA en Californie, il rencontre Iordanis Kerenidis (CNRS),\u00a0son futur directeur de th\u00e8se. Ils d\u00e9cident\u00a0de commencer une th\u00e8se \u00e0 l’Universit\u00e9 Paris Cit\u00e9\u00a0sur le sujet des algorithmes quantiques en intelligence artificielle \u00e0 l’IRIF (Institut de Recherche en Informatique Fondamentale). L\u2019ordinateur quantique nourrit de nombreux espoirs, mais de l\u2019incertitude r\u00e9side quant \u00e0 ses applications. Dans cette th\u00e8se, nous voulons savoir si les algorithmes quantiques peuvent \u00eatre utilis\u00e9s pour l\u2019apprentissage automatique. Nous montrons que des liens existent entre les deux concepts, \u00e0 travers l\u2019alg\u00e8bre lin\u00e9aire qui d\u00e9crit autant les \u00e9tats quantiques que les outils d\u2019apprentissage. Avec l\u2019analyse de la complexit\u00e9 et les simulations num\u00e9riques, nous prouvons que nos algorithmes quantiques sont plus rapides que leur version classique, malgr\u00e9 leur nature al\u00e9atoire. Nos algorithmes incluent les produits matriciels, l’estimation de distances, la d\u00e9composition de graphes, l’apprentissage non supervis\u00e9 (k-means, spectral clustering), mais aussi les r\u00e9seaux de neurones artificiels gr\u00e2ce au produit de convolution quantique pour images et une nouvelle m\u00e9thode de tomographie. Enfin, un circuit pour r\u00e9seaux neuronaux orthogonaux adapt\u00e9 aux premiers ordinateurs quantiques a \u00e9t\u00e9 exp\u00e9riment\u00e9.<\/p>\n

Les techniques chinoises occup\u00e8rent une place singuli\u00e8re dans la pens\u00e9e politique, \u00e9conomique et savante du XVIIIe si\u00e8cle en France. Un ensemble de facteurs convergents en Chine et en Europe (religieux, commerciaux, savants, politiques) contribu\u00e8rent \u00e0 faire de l’empire des Ming et surtout des Qing un mod\u00e8le d’expertise en mati\u00e8re de techniques dans un champ \u00e9largi de savoir-faire. Le royaume de France, en raison du r\u00f4le strat\u00e9gique qu’occupa la mission j\u00e9suite de P\u00e9kin tout au long du XVIIIe<\/sup> si\u00e8cle et des liens tiss\u00e9s entre \u00c9tat, science et entreprise depuis Colbert, fut aux avant-postes de circulations majeures d’informations sur les techniques chinoises. Apr\u00e8s un cursus en classe pr\u00e9paratoire aux grandes \u00e9coles, Ariles Remaki a suivi des \u00e9tudes de math\u00e9matiques \u00e0 l\u2019Ecole Normale Sup\u00e9rieure de Lyon et \u00e0 l\u2019Ecole Normale Sup\u00e9rieure de Cachan (actuelle ENS Paris-Saclay), entre 2012 et 2016. Il a alors eu l\u2019occasion de suivre quelques enseignements d\u2019histoire des math\u00e9matiques et des sciences en g\u00e9n\u00e9ral. Cela a provoqu\u00e9 chez lui un vif enthousiasme, en contentant l\u2019envie qu’ il avait d\u2019adopter une d\u00e9marche plus r\u00e9flexive vis-\u00e0-vis des savoirs auxquels il avait \u00e9t\u00e9 form\u00e9. Il a alors suivi le Master LoPHiSS-SPH d\u2019histoire et de philosophie des sciences au sein de l\u2019Universit\u00e9 Paris Diderot Paris 7 (actuelle Universit\u00e9 Paris Cit\u00e9). Sous la direction de David Rabouin, il a soutenu un m\u00e9moire en 2017 portant sur la Dissertatio de Arte Combinatoria<\/em>, ouvrage de jeunesse de G.W. Leibniz. Dans la continuit\u00e9 de ce travail, et toujours sous la direction de David Rabouin, il a d\u00e9but\u00e9 en septembre 2017 une th\u00e8se portant sur la combinatoire chez Leibniz.
En octobre 2019, il a \u00e9t\u00e9 laur\u00e9at d\u2019une bourse de la DAAD, gr\u00e2ce \u00e0 laquelle il a pu effectuer un s\u00e9jour de recherche de deux mois \u00e0 l\u2019Acad\u00e9mie de sciences de Berlin. Durant ce s\u00e9jour, il a pu collaborer avec le principal sp\u00e9cialiste de la combinatoire leibnizienne Eberhard Knobloch, ce qui a \u00e9t\u00e9 un apport extr\u00eamement pr\u00e9cieux et formateur pour son travail de recherche qu’il a soutenu en d\u00e9cembre 2021.
L\u2019\u00e9tude des manuscrits math\u00e9matiques de Leibniz a connu, cette derni\u00e8re d\u00e9cennie, un dynamisme sans pr\u00e9c\u00e9dent, auquel a fortement contribu\u00e9 l\u2019action scientifique de David Rabouin. En r\u00e9digeant sath\u00e8se de doctorat sous sa direction, Ariles a pu saisir l\u2019opportunit\u00e9 de s\u2019investir au sein de cette dynamique. Cela l\u2019a conduit \u00e0 int\u00e9grer le projet ERC men\u00e9 par David Rabouin, au sein duquel il est actuellement chercheur contractuel. Dans ce projet, il m\u00e8ne notamment une r\u00e9flexion sur les pratiques \u00e9ditoriales des textes math\u00e9matiques et l\u2019apport que peuvent pr\u00e9senter les outils num\u00e9riques au sein de telles entreprises.<\/p>\n

Ma th\u00e8se, intitul\u00e9e \u00ab L’art combinatoire comme art d’inventer chez Leibniz entre 1672 et 1680 \u00bb, porte sur le r\u00f4le architectonique de la combinatoire au sein des m\u00e9thodes de d\u00e9couvertes en math\u00e9matiques chez le philosophe allemand, principalement durant la p\u00e9riode 1672-1680. La combinatoire tient une place particuli\u00e8rement importante dans l\u2019\u0153uvre de Leibniz, mais son statut est ambigu et pose de s\u00e9rieuses difficult\u00e9s. En effet, suivant le contexte, la combinatoire peut d\u00e9signer aussi bien une m\u00e9thode d’investigation, un domaine de connaissances, un principe m\u00e9taphysique ou un outil de calcul. Mettre de l’ordre dans ces diff\u00e9rentes acceptions au travers de l’immense corpus leibnizien constitue un chantier consid\u00e9rable, dont ce travail examine un aspect qui avait \u00e9t\u00e9 n\u00e9glig\u00e9 jusqu’alors. Pour Leibinz, il y a un rapport \u00e9troit entre la combinatoire et l’art d’inventer, c’est-\u00e0-dire les m\u00e9thodes pour d\u00e9couvrir de nouvelles connaissances.<\/p>\n

Le discours th\u00e9orique et programmatique que tient le philosophe sur cette imbrication ont pouss\u00e9 les commentateurs, en particulier le logicien\u00a0Louis Couturat\u00a0\u00e0 porter l’attention sur son projet de \u00ab\u00a0caract\u00e9ristique universelle\u00a0\u00bb, autour de laquelle il d\u00e9veloppe plusieurs \u00e9bauches de syst\u00e8mes logiques et \u00e9pist\u00e9mologiques dans les ann\u00e9es 1677-1682. Ces observateurs se sont appuy\u00e9s sur ce corpus pour soutenir l’hypoth\u00e8se d’un \u00e9difice leibnizien unifi\u00e9 au sein duquel la combinatoire est assimil\u00e9e \u00e0 un syst\u00e8me logique. Les principaux \u00e9cueils auxquels m\u00e8ne cette interpr\u00e9tation sont d’ordre m\u00e9thodologique et historiographique.\u00a0<\/p>\n

Tout d’abord, l’id\u00e9e d’un syst\u00e8me philosophique coh\u00e9rent et homog\u00e8ne chez Leibniz est largement rejet\u00e9e par la majorit\u00e9 de la communaut\u00e9 scientifique actuelle.\u00a0R\u00e9tablir l’exigence historique, ce qui fut notamment permis par la forte croissance du mat\u00e9riel \u00e9dit\u00e9 disponible, constitue donc une premi\u00e8re \u00e9tape, \u00e0 laquelle s’est ajout\u00e9e une seconde : \u00e9largir et diversifier la conception de la combinatoire afin qu’elle s’accorde \u00e0 la r\u00e9alit\u00e9 pratique que recouvre ce terme au sein de l\u2019immense corpus.\u00a0
Mon travail de th\u00e8se a permis de mettre en \u00e9vidence trois acceptions principales de la combinatoire qui permettent de mieux comprendre son \u00e9volution dynamique et de dresser un premier paysage de son parcours durant la p\u00e9riode \u00e0 laquelle j\u2019ai restreint mon \u00e9tude. Je qualifie la premi\u00e8re acception de m\u00e9thodologique. Dans ce sens, la combinatoire est un synonyme de la synth\u00e8se et d\u00e9signe toute m\u00e9thode de d\u00e9couverte qui consiste \u00e0 adjoindre des \u00e9l\u00e9ments ext\u00e9rieurs aux donn\u00e9s afin de trouver de nouvelles choses. La seconde acception correspond \u00e0 ce que Leibniz nomme la science des formes et des formules, du semblable et du dissemblable. Je l\u2019appelle la combinatoire \u00e9pist\u00e9mologique, puisqu\u2019il s\u2019agit d\u2019un domaine du savoir \u00e0 part enti\u00e8re. Pour finir, la troisi\u00e8me acception d\u00e9signe la combinatoire dans son sens actuel au sein de math\u00e9matiques, \u00e0 savoir principalement une science du d\u00e9nombrement.\u00a0<\/p>\n

L’aspect purement math\u00e9matique a \u00e9t\u00e9 trait\u00e9 par l’historien des math\u00e9matiques Eberhard Knobloch, qui a rep\u00e9r\u00e9, comment\u00e9 et \u00e9dit\u00e9 la majeure partie des travaux en combinatoire stricto sensu chez Leibniz. Si l\u2019on adjoint \u00e0 ce corpus de combinatoire stricto-sensu, publi\u00e9 en 1976, celui qu\u2019il a publi\u00e9 en 1980 sur la th\u00e9orie des d\u00e9terminants, on obtient en r\u00e9alit\u00e9 un ensemble de texte propre \u00e0 \u00e9clairer une partie cons\u00e9quente de l\u2019\u00e9volution de l\u2019acception \u00e9pist\u00e9mologique. De plus, l\u2019\u00e9tude des premiers travaux de Leibniz en alg\u00e8bre, durant son s\u00e9jour parisien, est n\u00e9cessaire afin de saisir la mani\u00e8re dont la combinatoire \u00e9pist\u00e9mologique s\u2019est cristallis\u00e9e autour de la notion de forme.<\/p>\n

Cependant j\u2019ai fait le choix de laisser cette entreprise pour plus tard afin de consacrer l\u2019essentiel de mon travail de th\u00e8se \u00e0 la description de la premi\u00e8re acception de la combinatoire chez Leibniz, \u00e0 savoir la combinatoire m\u00e9thodologique. En effet, j\u2019ai d\u00e9couvert qu\u2019en laissant la focale sur la sciences de formes et sur les questions de caract\u00e9ristique, on est conduit \u00e0 manquer la premi\u00e8re occasion du mariage de la combinatoire et des math\u00e9matiques, \u00e0 savoir la pratique des tables num\u00e9riques.
Les tables et les diagrammes et la place qu\u2019ils ont au sein de la pratique math\u00e9matique est un sujet auquel Leibniz a beaucoup\u00a0r\u00e9fl\u00e9chi. Cependant, sur ce sujet comme sur d\u2019autres, la position de philosophe \u00e9volue au cours de sa vie : les diagrammes sont-ils des outils qui servent seulement l\u2019ars inveniendi ou bien peuvent-ils \u00eatre au services de l’ars judicandi\u00a0<\/em>?\u00a0Les diagrammes sont-ils des repr\u00e9sentations en relation avec notre imagination, ou bien des caract\u00e8res qui parlent \u00e0 notre entendement ? L\u2019\u00e9tude de la pratique concr\u00e8te des tables au sein des manuscrits math\u00e9matiques de Leibniz permet de fournir un \u00e9clairage nouveaux \u00e0 ces questions.<\/p>\n

Mais cette \u00e9tude demande de faire appel \u00e0 des m\u00e9thodes g\u00e9n\u00e9tiques qui sont relativement jeunes au sein des \u00e9tudes leibniziennes. La g\u00e9n\u00e9tique des figures diagrammatiques est d’ailleurs un domaine de recherche r\u00e9cent, m\u00eame au dehors du monde leibnizien. Ainsi, j\u2019ai pu apporter un nouvel \u00e9clairage sur les discours plus matures du philosophe au sein desquels il affirme que la synth\u00e8se est une partir n\u00e9cessaire de l\u2019art d\u2019inventer. Cette aspect synth\u00e9tique des m\u00e9thodes de d\u00e9couvertes n\u2019est pas seulement le fruit d’une op\u00e9ration mentale mais \u00e9galement le produit d\u2019une pratique, \u00e0 laquelle les tr\u00e8s nombreux brouillons math\u00e9matiques de Leibniz nous donne acc\u00e8s. En menant un \u00e9tude g\u00e9n\u00e9tique de ces proc\u00e9dures et en les inscrivant dans leur mat\u00e9rialit\u00e9 archivistique, j\u2019ai pu \u00e9galement mener une r\u00e9flexion sur le r\u00f4le de l\u2019exp\u00e9rience dans la pratique des math\u00e9matiques.<\/p>\n
